IMPORTANT: Avoid long term exposure to light and repeated freeze thaw cycles.
Procedure:
1. Thaw a tube of LysoTracker (1mM in DMSO).
2. Make a 1µM LysoTracker solution in 1 X dPBS by transferring 1µl of LysoTracker(1mM) into 1mL of 1 X dPBS.
3. Dissect gland and transfer gland into 2 well glass slide containing about 4-5 drops of LysoTracker solution.
4. Incubate gland at RT for about 2 minutes, then transfer gland into a 2 well glass slide containing fresh 1 X dPBS to wash.
5. Wash twice for about a minute.*
6. Prepare slide for microscopy as usual.
Notes
* Washing once for a minute doesn't seem to affect anything.
